Plant Protein vs Animal Protein: What Science Says About Muscle Growth
A meta-analysis of 43 RCTs (Nutrition Reviews, 2025) shows that animal protein slightly outperforms plant protein for muscle mass gain — SMD −0.20. But comparing soy to dairy reveals no difference. And adding leucine to plant protein closes the gap entirely.
According to a meta-analysis of 43 RCTs, animal protein provides a small advantage for muscle mass gain (SMD −0.20), but the difference is statistically non-significant in older adults and disappears entirely when soy is compared to dairy. The key factor is leucine: adding 1.5 g leucine to plant protein closes the gap with whey in muscle protein synthesis.
Why Is Leucine the Key Trigger for Muscle Protein Synthesis?
Muscle tissue growth is initiated through the mTORC1 signalling pathway. Leucine is an essential amino acid that activates this pathway directly. At least 2–3 g of leucine per meal is needed to trigger muscle protein synthesis after resistance exercise or food intake.
Leucine is where most of the difference between protein sources lies. Whey protein contains approximately 3 g of leucine per 20 g of protein. Pea, rice and wheat contain 1.5–2 g of leucine per 20 g of protein. Soy protein sits in between — around 2 g. This is not a bias against plants; it is amino acid arithmetic.
What Did the Acute Muscle Protein Synthesis Meta-Analysis Show?
A systematic review and meta-analysis of 12 studies (SportRxiv / American Journal of Clinical Nutrition, 2024–2025) compared acute muscle protein synthesis rates (fractional synthesis rate, FSR) after plant versus animal protein intake. Animal protein produced virtually the same synthesis rate: the between-group difference was 0.004 %/h (95% CrI: −0.002 to 0.011).
The gap was minimal: in young adults, differences in muscle protein synthesis after different protein sources were negligible. Nine of twelve studies (75%) found no statistically significant difference. Among older adults the picture differed slightly — animal protein showed a more pronounced effect, though data were heterogeneous even there.
What Did the Long-Term RCT Meta-Analysis on Muscle Mass Show?
The meta-analysis in Nutrition Reviews (2025) included 43 randomised controlled trials involving 1,538 participants. It is the most comprehensive analysis available on this topic. Main finding: animal protein slightly outperforms plant protein for muscle mass gain (SMD = −0.20; 95% CI: −0.37 to −0.03; p = 0.02).
However, a closer look at the data changes the picture:
- Soy protein vs dairy: no difference (17 RCTs; SMD = −0.02; p = 0.80). Soy is the exception among plant sources.
- Non-soy plant proteins (rice, chia, oat, potato) vs animal: larger gap (5 RCTs; SMD = −0.58; p = 0.02).
- Strength outcomes: no difference between protein sources (14 RCTs; p > 0.05).
- Physical performance: no difference (5 RCTs; p > 0.05).
- Older adults (60+): difference in muscle mass non-significant (SMD = −0.05; p = 0.74). Young adults are the only subgroup where animal protein advantage is significant (SMD = −0.28; p = 0.01).
What Happens When Leucine Is Added to Plant Protein?
A controlled crossover RCT published in Current Developments in Nutrition (2024) gave a direct answer to this question. Eight young healthy adults received on separate days: plant protein (20 g, 1.5 g leucine), plant protein + 1.5 g leucine (20 g protein, 3 g leucine total), whey protein (20 g, ~3 g leucine). Muscle protein synthesis rate was measured using isotope tracing.
Results:
- Plant protein: 0.041 %/h
- Plant protein + leucine: 0.049 %/h
- Whey protein: 0.046 %/h
Adding leucine increased synthesis by 16.2% compared to plant protein alone (p = 0.002). Whey protein outperformed plant protein without leucine by 12.1% (p = 0.046). The difference between plant + leucine and whey became statistically non-significant (p = 0.052). Authors' conclusion: leucine availability is the critical factor, not protein origin.
How Should Plant Protein Quality Be Assessed?
The standard for protein quality is the DIAAS (Digestible Indispensable Amino Acid Score), developed by the FAO. It accounts for both amino acid composition and digestibility. A score above 1.0 is considered "excellent", 0.75–0.99 "good".
Milk casein and egg white have DIAAS above 1.1. Soy protein isolate scores around 0.9–1.0. Pea protein isolate scores around 0.82. Wheat protein scores around 0.45. This means that wheat and rice deliver fewer digestible essential amino acids per gram of protein than dairy or eggs.
Practical takeaway: on a plant-based diet, diversifying protein sources (legumes plus grains, pea plus rice) raises the overall amino acid profile and compensates for the limiting amino acids in any single source.
- Protein source — animal or plant — matters less than adequate total dose and leucine content per meal (at least 2–3 g).
- Soy protein is comparable to dairy for muscle mass (SMD −0.02, non-significant). If you follow a plant-based diet, soy is the first priority.
- Non-soy plant proteins (pea, rice, wheat) fall behind animal sources — compensate by adding 1.5–2 g extra leucine per protein serving, or combine multiple sources.
- Strength outcomes do not depend on protein source: with the same training protocol and total protein intake, strength gains are equal.
- For older adults (60+), the difference in muscle mass between sources is non-significant — priority is total protein intake (1.6–2.2 g/kg/day), not origin.
- Assess quality by DIAAS, not by the "plant" or "animal" label. Pea protein isolate (DIAAS ~0.82) outperforms wheat protein (DIAAS ~0.45).
